Mazdutide (IBI362), a dual agonist targeting both GLP-1 and glucagon receptors, has shown promising results in clinical trials for weight management and metabolic disorders. While extensive human studies have been conducted, preclinical research, particularly in rat models, is limited. Nonetheless, available studies provide valuable insights into its potential applications.

In a study involving hyperuricemic rats, mazdutide administration significantly reduced blood uric acid levels, outperforming semaglutide and showing comparable efficacy to allopurinol, a standard treatment for hyperuricemia. Additionally, improvements in blood creatinine and urea nitrogen levels were observed, indicating potential benefits for kidney function (Innovent Biologics, 2023).

Another study demonstrated that mazdutide administration in diabetic mice led to improved glucose control and reduced body weight, even in genetically modified mice lacking either GLP-1 or glucagon receptors. This suggests that mazdutide’s effects may be mediated through both receptor pathways (Ji et al., 2022).

Furthermore, mazdutide has been associated with reductions in liver fat content and improvements in insulin sensitivity in clinical trials, highlighting its potential for treating non-alcoholic fatty liver disease and type 2 diabetes (Innovent Biologics, 2023).

While these studies provide a foundation for understanding mazdutide’s effects in animal models, further research is necessary to fully elucidate its mechanisms and therapeutic potential. The combination of GLP-1 and glucagon receptor activation presents a novel approach to metabolic disease management, warranting continued investigation in preclinical and clinical setting.
